How I can set/get current shortcut for my menu item?

How I can read/write value of any preference?

You can define a shortcut for a Plug-In menu function through Tools > Preferences > Key configuration.

No, the Plug-In cannot define the shortcut.

You plan to add these functions in future?

I have added this to the list of enhancement requests. Generally you do not want a Plug-In to override a user-defined function key though.
